Argatroban is a synthetic, direct thrombin inhibitor that provides anticoagulation through reversible, competitive binding to the active catalytic site of thrombin, the central enzyme in the coagulation cascade. Unlike indirect thrombin inhibitors such as heparin, which require antithrombin III as a cofactor, argatroban exerts its anticoagulant effect independently of endogenous cofactors, binding directly to thrombin with high affinity and specificity.

By occupying the active site of thrombin, argatroban prevents the cleavage of fibrinogen to fibrin, the activation of factors V, VIII, XI, and XIII, and the activation of platelets via protease-activated receptors.
