It exerts its anticoagulant effect by reversibly binding to the active catalytic site of thrombin, inhibiting both free and clot-bound thrombin, thereby preventing fibrin formation and platelet activation. Administered intravenously, argatroban has a predictable pharmacokinetic profile with rapid onset and short half-life, allowing for precise titration based on activated partial thromboplastin time (aPTT) monitoring.

Application & Function
Argatroban Application

Argatroban is a synthetic, direct thrombin inhibitor that provides anticoagulation through reversible, competitive binding to the active catalytic site of thrombin, the central enzyme in the coagulation cascade. Unlike indirect thrombin inhibitors such as heparin, which require antithrombin III as a cofactor, argatroban exerts its anticoagulant effect independently of endogenous cofactors, binding directly to thrombin with high affinity and specificity.

By occupying the active site of thrombin, argatroban prevents the cleavage of fibrinogen to fibrin, the activation of factors V, VIII, XI, and XIII, and the activation of platelets via protease-activated receptors. This inhibition is effective against both free (fluid-phase) thrombin and clot-bound thrombin.

Frequently Asked Questions
What is Argatroban used for?
Argatroban is used as an anticoagulant to prevent and treat thrombosis in patients with heparin-induced thrombocytopenia (HIT) and for those undergoing percutaneous coronary intervention (PCI).
How does Argatroban differ from heparin?
Unlike heparin, which requires antithrombin III as a cofactor, Argatroban is a direct thrombin inhibitor that binds directly to the active site of thrombin independently of endogenous cofactors.
What is the appearance of this product?
The product is a white to off-white powder, classified as pharmaceutical grade with an assay of 99%.
What are the storage requirements?
It should be stored in a ventilation drying environment to maintain its stability and quality.
What is the shelf life of Argatroban powder?
The shelf life of the Argatroban powder is 2 years when stored under proper conditions.
Does Argatroban inhibit clot-bound thrombin?
Yes, Argatroban is effective against both free (fluid-phase) thrombin and clot-bound thrombin, providing a significant advantage in managing thrombotic risks.
